If the implication there is that Buffy is the first Slayer to have sex then Robin Wood has some serious questions to ask his mother next time he see's her.

Or is it that she's the first to have had sex with a vampire?

Also, how on earth is Giles KNOWING all this stuff, even by Buffy standards it's an infodump of (literally) cosmic proportions.
